Role of respiratory uncoupling in drug-induced mitochondrial permeability transition

چکیده
Mitochondrial injury contributes to severe drug-induced liver injury. Particularly, mitochondrial permeability transition (MPT) is thought be relevant cytolytic hepatitis. However, the mechanism of MPT unclear and prediction not adequately evaluated in preclinical stage. In a previous study, we found that troglitazone, drug withdrawn due injury, induced via mild depolarization probably resulting from uncoupling. Herein, investigated whether other drugs induce share similar properties as using isolated mitochondria rat liver. Of 22 test examined, six drugs, including showed an uncoupling effect. Additionally, receiver operating characteristic analysis was conducted predict potential respiratory control ratio, indicator intensity. Results 2.5 best threshold exhibited high sensitivity (1.00) specificity (0.81), indicating correlated with potential. Activation calcium-independent phospholipase A2 appeared involved uncoupling-induced MPT. Furthermore, strong relationship between intensity effect among compounds confirmed. These results may help predicting cultured cells modifying chemical structures reduce risk. • The correlates Uncoupling induces iPLA 2 activation. Structure-activity for applicable

The role of reactive oxygen species in mitochondrial permeability transition.
We have provided evidence that mitochondrial membrane permeability transition induced by inorganic phosphate, uncouplers or prooxidants such as t-butyl hydroperoxide and diamide is caused by a Ca(2+)-stimulated production of reactive oxygen species (ROS) by the respiratory chain, at the level of the coenzyme Q. The ROS attack to membrane protein thiols produces cross-linkage reactions, that may...
متن کاملRole of the mitochondrial permeability transition pore in apoptosis.
Mitochondrial permeability transition (PT) involves the formation of proteaceous, regulated pores, probably by apposition of inner and outer mitochondrial membrane proteins which cooperate to form the mitochondrial megachannel (= mitochondrial PT pore). PT has important metabolic consequences, namely the collapse of the mitochondrial transmembrane potential, uncoupling of the respiratory chain,...
متن کاملRole of mitochondrial permeability transition pore in coated-platelet formation.
OBJECTIVE Coated-platelets are a subset of cells observed during costimulation of platelets with collagen and thrombin. Important characteristics of coated-platelets include retention of multiple alpha-granule proteins and expression of phosphatidylserine on the cell surface. The mitochondrial permeability transition pore (MPTP) is a key step in apoptosis and is suggested to be involved in some...
متن کاملRole of the mitochondrial permeability transition in myocardial disease.
Mitochondria play a key role in determining cell fate during exposure to stress. Their role during ischemia/reperfusion is particularly critical because of the conditions that promote both apoptosis by the mitochondrial pathway and necrosis by irreversible damage to mitochondria in association with mitochondrial permeability transition (MPT). MPT is caused by the opening of permeability transit...

42 Onset of the mitochondrial permeability transition (MPT) is the penultimate event 43 leading to lethal cellular ischemia/reperfusion injury, but the mechanisms precipitating 44 the MPT after reperfusion remain unclear. Here, we investigated the role of 45 mitochondrial free Ca and reactive oxygen species (ROS) in pHand MPT-dependent 46 reperfusion injury to hepatocytes. Cultured rat hepatocy...
